The role of sprouted fenugreek seeds as a nutraceutical in the protection against rats' cisplatin-induced nephrotoxicity and hepatotoxicity

Abstract

Germinated fenugreek seeds are plentiful in bioactive antioxidant used as a vital ingredient .Cisplatin (Cis) is an antineoplastic factor used for treating a variety types of cancer,however, it has severe toxic effects that interfere with its therapeutic efficiency. The present study was performed to better understanding the role of sprouted fenugreek seeds in the protection against Cis-induced nephrotoxicity and hepatotoxicity in rats. The experimental protocol designed as the animals(24 rats) were divided into four groups (six rats each); group (1) negative control group was fed on basal diet only, groups(2,3,4) were injected interaperitoneal by cisplatin (5mg/kg body wt.) from the first day and waiting for 24 hours to induce nephrotoxicity and hepatotoxicity . Positive control group (group 2) was fed on basal diet after injection, group(3,4) was fed basal diet +5% and 10% sprouted fenugreek seeds respectively after injection.The experiment continues for 28 days. Biochemical analysis in the liver and kidney were performed in addition to histopathological examinations.Cisplatin treatment increased creatinine,urea,uric acid,malondialdehyde (MDA) in the kidney, and declined the total antioxidant capacity(TAC).Also, the depletion was noticed in glutathione(GSH) as well as the reduction in the activities of the livar superoxide dismutase (SOD),catalase(CAT) in addition to increasing the albumin (Alb), total protein, alanine aminotransferase(ALT),and spartate aminotransferase (AST).The treatment with (5% and 10% sprouted fenugreek seeds) showed replenishment of antioxidant status and brought all the values to the levels close to normal. Phytochemicals present in fenugreek play an important role in ameliorating the Cis- toxicity in the liver and kidney.
